Ethanol Production in Actinomycetes after Expression of Synthetic adhB and pdc

چکیده
Streptomyces strains are producing many important secondary metabolites, in many cases used as pharmaceutical drugs. Though the usage of these bacteria for the production of fuel has never been described. We could show, that the expression of two artificial ethanologenic genes of Zymomonas mobilis in different actinomycetes strains resulted in the production of ethanol. The synthetic genes adhB and pdc encoding an alcohol dehydrogenase and a pyruvate decarboxylase, respectively, were expressed in eight different Streptomyces strains. Best production was obtained using Streptomyces coelicolor A3(2) harboring both genes. By variation of the cultivation conditions, the amount of ethanol produced by the strain could be increased up to 2.6 g/l.

The optimization of synthetic pathways is a central challenge in metabolic engineering. OptSSeq (Optimization by Selection and Sequencing) is one approach to this challenge. OptSSeq couples selection of optimal enzyme expression levels linked to cell growth rate with high-throughput sequencing to track enrichment of gene expression elements (promoters and ribosome-binding sites) from a combinat...
متن کاملThermotolerant Zymomonas mobilis: Comparison of Ethanol Fermenta- tion Capability with that of an Efficient Type Strain
Zymomonas mobilis is an alternative microorganism to Saccharomyces cerevisiae for ethanol production. To find a thermotolerant Z. mobilis strain, the growth and ethanol production of four isolates in Thailand were compared with those of the efficient strain ZM4 (NRRL B-14023) at different temperatures. One of the selected strains, TISTR 405, was found to grow and produce ethanol even at 39 ̊C to...

Lactic acid bacteria (LAB) are known for their high tolerance toward organic acids and alcohols (R. S. Gold, M. M. Meagher, R. Hutkins, and T. Conway, J. Ind. Microbiol. 10:45-54, 1992) and could potentially serve as platform organisms for production of these compounds. In this study, we attempted to redirect the metabolism of LAB model organism Lactococcus lactis toward ethanol production. Cod...

Wild-type Bacillus subtilis ferments 20 g/liter glucose in 48 h, producing lactate and butanediol, but not ethanol or acetate. To construct an ethanologenic B. subtilis strain, homologous recombination was used to disrupt the native lactate dehydrogenase (LDH) gene (ldh) by chromosomal insertion of the Zymomonas mobilis pyruvate decarboxylase gene (pdc) and alcohol dehydrogenase II gene (adhB) ...

BACKGROUND Pyruvate decarboxylase (PDC) is a well-known pathway for ethanol production, but has not been demonstrated for high titer ethanol production at temperatures above 50 °C. RESULT Here we examined the thermostability of eight PDCs. The purified bacterial enzymes retained 20% of activity after incubation for 30 min at 55 °C. Expression of these PDC genes, except the one from Zymomonas ...
